An endonuclease that acts on phosphodiester linkages, particularly linkages adjacent to pyrimidine nucleosides, resulting in polynucleotides with a free hydroxyl group at the 3' end and a phosphate at the 5' end. The optimum pH for enzymatic action is 7.8. DNase can be activated by divalent metals and inhibited by Chemicalbook chelates such as EDTA and sodium lauryl sulfate. Calcium ions at a concentration of 5 mM act as a stabilizer to protect DNase from being broken down by hydrolases. It is mainly used to degrade DNA in RNA, purify RNA, and study the interaction between DNA and protein.
